Engineering Saccharomyces cerevisiae for Increased Utilization of Xylose from Biomass

ثبت نشده
چکیده

What problem does it address? Biomass feedstocks are composed of hexose and pentose sugars locked into a higher-order structure. The most abundant of these sugars are glucose and xylose. Strains have been engineered to utilize xylose, but these strains do not grow well when xylose is the only sugar available. Xylose uptake into the cell is one of the limitations to efficient xylose utilization. The strain developed here addresses the problem of poor xylose uptake in S. cerevisiae.

Evolutionary engineering of Saccharomyces cerevisiae for efficient aerobic xylose consumption.

Industrial biotechnology aims to develop robust microbial cell factories, such as Saccharomyces cerevisiae, to produce an array of added value chemicals presently dominated by petrochemical processes. Xylose is the second most abundant monosaccharide after glucose and the most prevalent pentose sugar found in lignocelluloses. Significant research efforts have focused on the metabolic engineerin...

Increased ethanol productivity in xylose-utilizing Saccharomyces cerevisiae via a randomly mutagenized xylose reductase.

Baker's yeast (Saccharomyces cerevisiae) has been genetically engineered to ferment the pentose sugar xylose present in lignocellulose biomass. One of the reactions controlling the rate of xylose utilization is catalyzed by xylose reductase (XR). In particular, the cofactor specificity of XR is not optimized with respect to the downstream pathway, and the reaction rate is insufficient for high ...

Metabolic Engineering of Yeast for Xylose Uptake and Fermentation

Fuel ethanol production from plant biomass hydrolysates by the baker’s yeast Saccharomyces cerevisiae is of great economic and environmental significance. Costeffective production of ethanol from plant biomass should not only be based on the readily fermentable starch and sucrose fractions of plant carbohydrates, but also on the much more resistant lignocellulosic fractions. The utilization of ...

Bulk Segregant Analysis by High-Throughput Sequencing Reveals a Novel Xylose Utilization Gene from Saccharomyces cerevisiae

Fermentation of xylose is a fundamental requirement for the efficient production of ethanol from lignocellulosic biomass sources. Although they aggressively ferment hexoses, it has long been thought that native Saccharomyces cerevisiae strains cannot grow fermentatively or non-fermentatively on xylose. Population surveys have uncovered a few naturally occurring strains that are weakly xylose-po...

Evolved hexose transporter enhances xylose uptake and glucose/xylose co-utilization in Saccharomyces cerevisiae

Enhancing xylose utilization has been a major focus in Saccharomyces cerevisiae strain-engineering efforts. The incentive for these studies arises from the need to use all sugars in the typical carbon mixtures that comprise standard renewable plant-biomass-based carbon sources. While major advances have been made in developing utilization pathways, the efficient import of five carbon sugars int...
